Organization of the human KIR locus

Killer cell immunoglobulin-like receptors (KIR) play a role in regulating the functional activity of Natural Killer (NK) cells. Expressed on the surface of NK cells and some T cells, they recognize HLA Class I molecules found on the surface of target cells. These receptors can be either inhibitory or activating in nature.
 
In haploidentical hematopoietic stem cell transplantation, there is evidence that when the recipient lacks the inhibitory HLA ligand for the donor’s NK cells, there can be an NK-cell alloreaction. This reaction can be in the GVL direction and can also help prevent GVHD.
